The possible meaning of the number 456 is derived primarily from its relationship to Abraham's family and the occurrence of certain words.
Entry 456 in the Hebrew definition portion of Strong's Concordance is eliychoreph. This word means the "God of autumn." It is used only once, in 1Kings 4:3, to refer to a man named Elihoreph who was one of the princes of Israel at the time of King Solomon.
So king Solomon was king over all Israel. And these were the princes which he had; Azariah the son of Zadok the priest, Elihoreph (eliychoreph) and Ahiah . . . (1Kings 4:1 - 3, KJV).
Entry 456 in the Greek definition portion of Strong's Concordance is anoikodomeo. The word means to rebuild or build again. It is found only in a single verse where it is utilized twice.
After this I will return, and will build again (anoikodomeo) the tabernacle of David, which is fallen down; and I will build again (anoikodomeo) the ruins thereof, and I will set it up (Acts 15:16, KJV).

The Gematria for Isaac (Strong's #H3327), the child of promise given by God to Abraham and Sarah in their old age, is 208 (10 + 90 + 8 + 100). Added together they equal 456.
The Hebrew Gematria for Jacob's name (Strong's #3290) is 182 (10 + 70 + 100 + 2). The Gematria for Leah's name (Strong's #H3812) is 36 (30 + 1 + 5). The Gematria for Rachel (#H7354), Jacob's other wife whom he loved the most, is 238 (200 + 8 + 30). All together, they add to 456.
